When the fateful letter arrives on the day of the planned trip to Whitwell, Mrs. Jennings asks of Colonel Brandon "Was it from Avignon? I hope it is not to say that your sister is worse." Based on this information, we can conclude that this sister would also have been the daughter of Mr. Brandon Senior, the sister of the elder Brandon son and the cousin of Whitwell in the first place, however, because it was owned by Brandon’s brother-in-law. It is unclear if this man is married to the sister in Avignon or if there are two different sisters. Given that Brandon could probably have sent a letter of introduction along if his sister was indeed in residence instead of cancelling the entire planned outing, it seems likely that it is the same sister in both instances.
